Test Code LAB744 Pyruvate

Important Note

Place the sample in a DRY bag, then place this bag into a bag of ice and send to the lab within 5 min. of collection. Note whether sample is venous or arterial (venous is preferable unless an arterial line is in place).

Deliver to CORE within 2 min of receipt in Lab. A protein-free filtrate must be made by CORE as soon as possible.

Clinical System Name

Pyruvate

Sample Requirements

Specimen: Whole blood

Container(s): Dark Green/Sodium Heparin, Gray/Sodium Fluoride, Heparinized Syringe

Preferred Vol: 1 mL

Minimum Vol: 1 mL

Note: Place the sample in a DRY bag, then place this bag into a bag of ice and send to the lab within 5 min. of collection. Note whether sample is venous or arterial (venous is preferable unless an arterial line is in place). Note also if the patient was crying or it was a difficult draw.

Processing Instructions

Reject due to: Specimens received not on ice, age of specimen, insufficient volume.

Spin: N

Aliquot: N

Temp: - 20 C

Storage location: Deliver to CORE within 2 min of receipt. A protein-free filtrate must be made by CORE as soon as possible.

CORE: Add 0.5 mL well-mixed whole blood to 1.0 mL cold 6% perchloric acid. Immediately mix by vortexing until the sample is chocolate brown. Centrifuge sample and aliquot the clear supernatant into a 4ml tube and labeled with the large Epic label.  Place aliquot in the "return to CPA" box.

CPA: Retrieve clear supernatant aliquot from CORE  and place in CPA freezer, Fz5 rack.

Off-site collection:  Draw at least 1 mL blood in a Dark Green/Sodium Heparin or Gray/Sodium Fluoride, no serum separator. Note if patient was crying or draw was difficult. Prepare protein-free filtrate: Add 0.5 mL well-mixed whole blood to 1.0 mL cold 6% perchloric acid (PCA). Mix by vortexing until the sample is a chocolate brown color. Centrifuge and transfer supernate to plastic tube.  Send frozen.

If PCA is unavailable, send whole blood in a gray top tube on ice within 2 hours.
